The overall goal of this project is to capitalize on proteins scaffolds for discovery, engineering, and production of pharmaceutical and agricultural agents. The project focuses on the antibacterial activity of a particular family of proteins that are expressed by certain plant species.These proteins, the cyclotides, have an extraordinary structure in that they are head-to-tail cyclic. Furthermore, they contain three disulfide bonds forming a cystine knot. Together these features renders them an extraordinary stability.In the plant, cyclotides form a part of the host defense system, and we have now discovered that we can use that defense against pathogenic bacteria. With the background of the increasing need of new antibiotics, the current project aims at revealing the structure activitty relationship of the antibacterial effects of cyclotides, their mode of action, and their biosynthesis, with the ultimate goal to set the stage for the design of an optimized cyclotide protein and for the expression of that cycloid using recombinant plants.To that end, peptide synthesis and structural studies, computer based structure-activity evaluations, antimicrobial and biophysical bioassays, and production of the cyclotide precursor protein are key elements.
