The effect of titanium dioxide nanoparticles on human ARPE-19 cells : A potential risk factor for age-related macular degeneration (English)
Abstract [sv]
Abstract [en]
Aim: The aim of the thesis was to investigate how human RPE cells were affected during long-term exposure to titanium dioxide nanoparticles (TiO2 -NP) and to investigate whether it may constitute a potential risk factor for age-related macular degeneration.
Method: ARPE-19 cells were cultured In Vitro and exposed to five different concentrations of TiO2 -NP in sizes of ≤ 21 nm and ≤ 100 nm over a period of three weeks. Two different controls were used, consisting of NaCl and cell culture medium. The number of dead cells were calculated using a cell counter. To investigate if the cells secreted VEGF, ELISA was used. Microscopy was performed to examine if the cells expressed a marker for oxidative stress on the cell surface. The size of TiO2 -NP was analyzed using dynamic light scattering.
Results: The viability measurement showed statistically significant cell death in ARPE-19 cells after exposure to TiO2 -NP ≥ 500 mg/mL. A dose-dependent increase of TiO2 -NP resulted in a statistically significant increase in cell death. Exposure to TiO2 -NP showed a clear impact on angiogenesis, with an initial decrease in VEGF secretion followed by recovery at higher TiO2 -NP concentrations. Oxidative stress was presented at all concentrations for ≤ 21 nm and ≤ 100 nm TiO2 -NP. Dynamic light scattering showed that TiO2 -NP aggregated at concentrations ≥ 5 mg/mL.
Conclusion: TiO2 -NP exposure induced increased cell death and oxidative stress in ARPE-19 cells and affected angiogenesis. To be able to draw more definitive conclusions, further and more extensive research is required, especially In Vivo and at realistic exposure levels. However, our In Vitro results indicate that TiO2 -NP may be a potential risk factor for the development of AMD.
